1)翻訳後修飾酵素を用いるタンパク質の部位特異的修飾法

概要:ペプチドやタンパク質中に含まれるグルタミン残基とリジン残基の間を選択的に架橋化する酵素トランスグルタミナーゼを使って、組換えタンパク質のペプチドタグを狙った部位特異的なタンパク質間架橋による二量体化、核酸-タンパク質コンジュゲートの調製等について研究を進めています。

3)生体触媒の高度利用によるバイオプロダクション

概要:昨今のエネルギー問題の解決には、生体触媒の高度利用ならびにバイオマスの利活用が鍵を握ると言われています。バイオマスの分解・変換におけるキープレーヤーである酵素を実用的な触媒として利用するべく、生化学(タンパク質工学)、有機化学、生物化学工学(酵素工学)をベースに、様々な角度から検討を行っています。

4)生物機能を模倣・利用するバイオアセンブリ

概要:自己組織化は、生命活動の様々な場面で普遍的に観察される現象です。当研究室では、特に、界面活性能を有する分子が自己組織的に形成するミセル(水中)あるいは逆ミセルやエマルジョン(非水溶媒中)のバイオ分野への応用について検討しています。前者について、最近、牛乳に大量に含まれるカゼインを化学的に架橋化処理することで、直径約20 nm程度の温度応答性タンパク質性ナノ粒子が簡単に得られることを発見しました。

5)薬理活性ペプチドやタンパク質の経皮デリバリー

概要:最近、薬理活性を有するペプチドや、抗体の様なタンパク質、さらにはsiRNA等のオリゴ核酸が、バイオ医薬として市場に台頭してきました。これらのバイオ医薬は、経口投与されると胃腸で分解されてしまうので、一般的に注射により投与されます。当研究グループでは、バイオ医薬の「塗り薬化」を目指しています。
